A-Alpha Bio is a biotechnology company using synthetic biology and machine learning to measure, discover, predict, and engineer protein-protein interactions. We pursue our mission of improving human health through an internal therapeutics pipeline and by partnering with industry-leading pharmaceutical and technology companies.

Current approaches for measuring protein-protein interactions are insufficient for many drug development applications due to low accuracy or throughput. We are a team of synthetic biologists, structural biologists, and next-generation sequencing experts who have come together to remove this industry-wide bottleneck. Our proprietary experimental platform, AlphaSeq, generates high-resolution data to discover rare and effective antibodies, identify degradable neo-substrates for molecular glues, and generate massive protein-protein interaction datasets to map the protein binding landscape. Our AlphaBind platform then leverages these data to train predictive machine learning models to further accelerate the discovery and optimization of high-impact therapeutics.

We are excited to share the results of our recent collaboration with Google and Lumen Bioscience! 

With funding from the Bill & Melinda Gates Foundation, we’ve used AlphaSeq and machine learning (ML) to generate highly diverse single-domain antibodies that bind and neutralize SARS-CoV-2 variants with broad cross-reactivity. 

Through multiple iterations of AlphaSeq data generation, ML model training, and antibody design, we generated thousands of antibody variants showing enhanced binding against diverse SARS-CoV-2 strains. The most promising antibody candidates were expressed recombinantly and evaluated with biolayer interferometry and viral neutralization assays, demonstrating up to 100-fold improvement in binding and neutralization compared to the parental antibody.

The end result is twofold: (1) therapeutic variants showing potent neutralization of diverse viral variants, and (2) an ML approach that can predict binding across vast regions of sequence space and may help future-proof therapeutics against rapidly evolving pathogens.
