Still buzzing after attending last week's Federation of American Societies for Experimental Biology (FASEB) conference on Genome Engineering held in the beautiful city of Rome. 

The conference was a treasure trove of knowledge, featuring incredible advancements in genome engineering. We heard about the latest developments in base and prime editing, novel CRISPR systems, precise therapeutic editing, and exciting progress in delivery methods to humans. The discussions were intellectually stimulating, with a key focus on:

1) **Overcoming unintended double-strand breaks (DSBs)** in our editing systems and developing methods to detect and quantify these DSBs accurately (check out what we do at Broken String Biosciences to address this problem!).

2) **Streamlining the preclinical drug development path** from both experimental and regulatory perspectives. This is crucial to ensuring safety while shortening development timelines and reducing costs, ultimately providing treatments for rare diseases and neglected patient populations (it was humbling to finally meet Fyodor Urnov, who has long been at the forefront of this endeavor 💫).

3) **Improving access to novel therapies**, including addressing cost barriers and ensuring access to clinical trials and/or marketed therapies. One additional recurring theme was the bias around reference genomes and the need to better understand genomic variations in different geographic regions and populations, which could impact the efficacy and safety of cell and gene therapies.

What made this event truly special was the vibrant community of scientists who gathered to share their insights and foster collaborations. The social interactions and community building were as enriching as the scientific content (obviously we had to sample gelato while in Rome!). The backdrop of Rome added a unique charm, providing the perfect setting for networking and forming lasting connections - combined by flawless organisation by Faseb's Joanna U. Engstrom, DES and team AND the scientific organisers Shengdar Q. Tsai, Nicole M. Gaudelli, Philip Gregory, Angelo Lombardo! 👏 👏

On a warm Sunday evening, David R. Liu kicked off an exceptionally exciting program for this year’s FASEB conference on Genome Engineering in the beautiful city of Rome 🇮🇹 
He presented his latest groundbreaking findings, called eePASSIGE—an improved version of PASSIGE (prime-editing-assisted site-specific integrase gene editing). This method, once the challenge of targeted and efficient delivery to humans is solved, could address a fundamental issue in the gene therapy field: how to insert a large piece of DNA into a specific genomic location.

In addition to David Liu's presentation, this meeting's agenda is packed with thrilling talks about novel advancements in genome engineering, ranging from base and prime editors to new CRISPR systems, precise therapeutic editing, and advancements in delivering these reagents to humans.

One major question being raised by both speakers and the audience alike is: How can we overcome unintended double-strand breaks (DSBs) in our editing systems, AND, if we cannot, how can we precisely and efficiently detect and quantify these wanted and unwanted DSBs? At Broken String Biosciences we have developed INDUCE-seq, a novel DSB assay that detects and quantifies DSBs in any cell type with unprecedented sensitivity.
