Why use HT parallel screening for API production? One of our clients needed a 300 g synthesis of an API for a toxicology study. The existing synthetic route had previously been achieved on 50 g scale but improvements to the key Suzuki coupling step were required to enable scale up. Use of our high-throughput parallel screening workflow uncovered a ligand that switched off the protodeborylation pathway reducing the amount of boronic acid required to one equivalent and which was accompanied by an increase in reaction yield. Within four weeks of development, the Suzuki reaction was successfully run on a 500 g scale and the product was rapidly converted to > 300 g of API and delivered to our client for their tox study. Most early-stage routes to an API are developed at speed for target screening. Consequently, many of these routes are developed to prepare milligrams of API and are therefore ill-suited to deliver enough material for IND-enabling studies. We can support the development of fit-for-purpose synthetic routes by identifying the problematic transformations and providing bespoke solutions.
