An international group, including researchers at the Wyss Institute at Harvard University for Biologically Inspired Engineering, Dana-Farber Cancer Institute, and Boston Children's Hospital, extended DoriVac, a #DNA origami-based #vaccine platform, to infectious diseases.
 
By conjugating DoriVac with heptad repeat 2 (HR2) peptides, which play a role in fusing the viral and host cell membranes and are conserved across a host of viral species, the group observed that DoriVac induced neutralizing antibodies and CD4+ and CD8+ T-cell response in mice against SARS-CoV-2, HIV and Ebola. Immune response was also induced in a human lymph-node-on-chip model.
 
Taken together, the results suggest DoriVac, when conjugated to viral antigens, holds promise for inducing robust immune response and its flexible, modular nature may make it a good candidate system for responding to future global disease outbreaks.

A group at Dana-Farber Cancer Institute , along with researchers at the Broad Institute of MIT and Harvard,  and Massachusetts General Hospital Cancer Center, previously reported on a new technique called MAESTRO (Minor Allele Enriched Sequencing Through Recognition Oligonucleotides) for assaying for minimal residual disease (#MRD) using #cfDNA. 
 
In a new study, this group expanded on MAESTRO with a new approach called MAESTRO-Pool. This technique pools MAESTRO probes from all patients assayed, which allowed the researchers to assay over 23,000 mutations in 9 melanoma patients over several cfDNA samples per patient. By pooling, the researchers were able to quantify MRD in matched patients as well as the rate of false positives in unmatched patients.
 
The assay showed a 10- to 100-fold improvement in sensitivity versus existing MRD tests and showed high specificity for detecting MRD.

As we bring 2023 to a close, we reflect on this past year and the promise of benchtop enzymatic #DNA synthesis (EDS), and look back at a publication by a group at US Army which assessed benchtop #EDS for gene assembly applications. 
 
The study concluded that EDS-derived #oligonucleotides could be obtained in half the time of commercially-synthesized oligos, yielded functional gene product, and that EDS clearly reduced the waste stream compared to using traditional chemical synthesis methods for oligo production.
