Yesterday's miniCUP Boston event brought together scientists, industry leaders, and researchers to discuss the latest advancements in computational chemistry and molecular modeling. Hot off the press! Cryptic pockets have the potential to open new therapeutic opportunities by revealing ligand binding sites that remain hidden in static apo structures of a target protein. To uncover these cryptic pockets, OpenEye’s scientists applied Weighted Ensemble path sampling to enhance protein motion along normal modes, and then used a combination of analysis methods—Exposon Analysis, Probe Map, and Dynamic Probe Binding—to successfully identify cryptic pockets on the G12D isoform of the KRAS oncoprotein.
