Using Farcast's TruTumor technology, tumors from 6 HNSCC patients were simultaneously treated with Nivolumab and Pembrolizumab. The Tumor Microdynamic response was measured and compared using multi-omic assays.

Tumors constantly shape their micro-environment by dynamic  signaling and communication with immune cells and constantly alter the immune and extracellular milieu. This allows tumor cells to create a tumor-supportive environment and induce neo-angiogenesis,  as well as activate a variety of mechanisms to hijack the micro-environment. 
Fighting cancer requires a deep understanding of tumor microdynamics and an intelligent decoding of myriad signals in every tumor, so that we can deliver durable benefits by directing appropriate therapies personalized for every tumor.

It is hard to anticipate the activation of antitumor immune response during immunotherapy. The success of any immunotherapy is multifactorial and results from complex interplay between tumor, immune and stromal microenvironments.

For instance, PD-1/PD-L1 blockade is associated with multiple functional attributes of immunity. While numerous biomarkers exist for multiple tumor types, an accurate identification of responder phenotype is still evasive.

Understanding tumor microdynamics is key to predicting response to immunotherapy.  When faced with failures like HPD,  use of combination therapies could come to rescue if one could ascertain the spatial and temporal intra-tumoral variability.
