Today, we shared results from the Phase 1 clinical trial of VENT-03, the first cGAS inhibitor to successfully complete a first-in-human study. In this trial, VENT-03 was safe and well-tolerated, showed a favorable PK profile that supports once-daily dosing, and demonstrated full target inhibition. These excellent results reinforce the significant potential of VENT-03 to be a first- and best-in-class oral anti-cGAS medication and emphasize the power of our ReSOLVE™ platform. It was an honor to be invited to present at the EMBO Inflammasome Workshop at Ludwig-Maximilians-Universität München in Munich. Our Chief Scientific Officer, Michael Crackower , presented our progress on the development of first-in-class caspase-4 inhibitors using our proprietary, biophysics-based and #machinelearning -enabled platform, ReSOLVE™. This is our first disclosure of our groundbreaking discovery of allosteric, selective, small-molecule caspase-4 inhibitors that are highly potent in vitro and in vivo and work via a previously undiscovered mechanism. The role of caspase-4 at the interface of bacterial components and the production of numerous cytokines makes this a highly compelling target for diseases of high unmet need like #InflammatoryBowelDisease , hidradenitis suppurative, and sepsis, where single cytokine inhibition has resulted in sub-optimal efficacy. We enjoyed an awesome summer outing in Downtown Boston, where cityHUNT Scavenger Hunt put our local knowledge to the test with a fun day of scavenger hunting.
