As experts in cytokine biology, our clinical stage pipeline comprises five cytokine-derived therapeutic candidates, with our lead proprietary compound, SON-1010, in Phase 1 development. Sonnet’s proprietary FHAB™ (Fully Human Albumin Binding) technology enables the development of innovative targeted biologic drugs with enhanced single- or bi-specific mechanisms. The FHAB™ platform utilizes a fully human single chain antibody fragment (scFv) that binds to and “hitchhikes” on human serum albumin (HSA) for transport to target tissues. Our internal focus is immune oncology, however, we believe our technology is suited for drug development across the spectrum of human disease.
 
Sonnet’s FHAB construct attaches to albumin in the bloodstream, which significantly enhances the pharmacokinetics of the active drug substance. It also naturally accumulates at inflammation sites, including tumors, thereby potentiating delivery to target tissues. Our platform has demonstrated a tenfold increase in half-life and a greater-than-thirtyfold increase in efficacy as compared to recombinant interleukins without FHAB in preclinical studies. We believe the versatile platform can generate a large immune oncology pipeline, including combinations with a variety of biologic drugs. The Sonnet platform is differentiated in that FHAB utilizes a fully human sequence with a human glycosylation profile that can be manufactured using a standard CHO process.

We’re excited to announce that we’ve been approved to begin a Phase 1b/2a clinical trial of SON-1010 in combo with Roche's atezolizumab (Tecentriq®) in Australia for patients with platinum-resistant ovarian cancer (PROC).

This combo trial is expected to show proof of concept with SON-1010 for PROC, as well as to augment the foundational work underway with Sonnet’s bifunctional molecules, SON-1210 and SON-1410.

Today we released our Q1 2023 earnings report, and it’s been an exciting period of progress for our company. Here are some key updates:

•At the American Association for Cancer Research Annual Meeting last month, we released new Phase 1 results for our lead candidate SON-1010 showing its potential as a best-in-class IL-12 cancer immunotherapy. The data reveals our candidate had a prolonged and controlled immune response without causing harmful side effects. And some patients with advanced solid tumors have shown clinical benefit.

•We recently entered into a collaboration agreement with Roche for the clinical evaluation of SON-1010 with atezolizumab (Tecentriq®), a checkpoint inhibitor, to treat patients with ovarian cancer.

•We announced positive results from safety studies of SON-1210 in non-human primates, showing that the drug candidate is safe even at high doses.

•We completed a $15 million financing to support the development of our pipeline and realize the potential of our FHAB (Fully Human Albumin Binding) technology.

We are very pleased to unveil our latest data from our lead candidate, SON-1010, showing not only positive safety and tolerability data, but early signs of potential clinical benefit in our cancer patient group. In our healthy volunteer group, SON-1010 exhibited extended pharmacokinetics (PK), which aligns with our preclinical models that show that the candidate drug can persist in the body to produce a therapeutic effect. In addition, in our cancer patient study group SON-1010 showed positive signs of tumor shrinkage. 

We’re thrilled with this progress and the early evidence to support SON-1010 as a best-in-class targeted cancer therapy.

We are excited to announce that SON-1210, our leading bifunctional candidate targeting solid tumors, has successfully concluded toxicology studies. This critical milestone enables us to begin regulatory filings during the first half of 2023 for SON-1210 to enter first-in-human clinical trials.

Developed using our Fully Human Albumin Binding (FHAB) technology platform, SON-1210 combines two cytokine drug candidates — human Interleukin 12 and Interleukin 15 — in a single molecule. The FHAB technology is designed to help cancer drugs more safely and effectively target tumors. And as a bifunctional therapeutic candidate, SON-1210 was created to deliver the two cytokines to local tumor tissue, helping to turn “cold” tumors “hot” to activate an anti-cancer immune response.

We designed our proprietary Fully Human Albumin Binding (FHAB) platform to overcome some of the major challenges in cytokine-based #immunotherapies. Historically, therapeutic cytokines don’t stay in the body long enough and can have toxic side effects. 

Now we’re excited to announce recent results from our phase 1 dose-escalation study of SON-1010 demonstrating the potential of our FHAB technology to overcome these cytokine delivery challenges in healthy humans. 

The recent PK/PD data shows that SON-1010 has an extended half-life of approximately 112 hours in healthy human volunteers and has an acceptable safety profile. Our technology is designed to be able to use higher doses of cytokines without triggering unacceptable toxicity — thereby activating an immune response to fight tumors. 

We look forward to upcoming collaborations to demonstrate the potential of this powerful cancer therapy.

For patients with platinum-resistant ovarian cancer no longer responding to chemotherapy, combination immunotherapies hold great promise. We’re excited to announce a collaboration with Roche to evaluate our lead product, SON-1010, in combination with Roche’s atezolizumab for patients with recurrent ovarian cancer. 

SON-1010 is a proprietary version of the immunotherapy drug interleukin-12 that’s designed to persist in the body longer and target tumors better. Atezolizumab is a commercially approved monoclonal antibody for the treatment of various solid tumor cancers.
