What if we could accurately simulate drug absorption and toxicity using a microphysiological system ( #MPS )? In an exciting new study, researchers from the Chinese Academy of Medical Sciences & Peking Union Medical College and TissUse GmbH (Berlin) have developed an intestinal-liver-on-chip model to evaluate hepatotoxicity after an acetaminophen (APAP) overdose. This innovative approach mimics drug absorption and metabolism, overcoming limitations seen in traditional animal models for drug safety evaluation. Hepatotoxic assessment in a micro physiological system: Simulation of the drug absorption and #toxic process after an overdosed acetaminophen on #intestinal -liver-on-chip Using Caco-2 and HT29-MTX-E12 cells to establish intestinal equivalents and HepG2, HUVEC-T1, PMA-induced THP-1, and hepatic stellate cells for liver equivalents, the study effectively simulated acute #liver injury post-APAP overdose. High-performance liquid chromatography was used to track APAP concentration, and various liver injury biomarkers, including aspartate aminotransferase (AST) and alanine aminotransferase (ALT), were monitored over 4 days, showing stable and reliable results. This #organ -on-chip model not only enhances our understanding of toxicokinetics but also provides a valuable tool for drug toxicity assays. The findings could further open doors for safer drug development with fewer animal models. We are thrilled to share our latest groundbreaking research with amazing collaborators from AstraZeneca and Linköping University —successfully replicating human glucose regulation in vitro using a pancreas-liver chip! This platform mimics the key interactions between these organs, enabling us to study glucocorticoid-induced diabetes while integrating mathematical modelling. Our results are reproducible across labs, giving us confidence that this model will unlock new therapeutic targets and transform drug discovery in metabolic disease research 🎯
